A falling-block puzzle game in which players combine four or more blocks of the same colour to make them disappear. The remaining blocks then fall, potentially creating combos that send special nuisance blocks to the opponent’s field; these can only be cleared by removing adjacent blocks. The game includes a story mode, in which Arle faces computer-controlled opponents, as well as a two-player versus mode, a four-player mode, and an endless mode.

Super Puyo Puyo 2 replaces the original game's linear campaign with a tower-climbing mode where Arle Nadja selects opponents by spinning a roulette. Each floor of the tower carries a required experience-point threshold that must be met to advance; if all available opponents are defeated without reaching the target, a hidden final challenger appears. Failing to hit the quota after that last match ends the run. Experience requirements vary between the arcade, Mega Drive, and Super Famicom releases—the Super Famicom version demanding twice the points on the first floor alone.
